A Method for Preparing Concentrates of Malarial Parasites from Chicks Infected with P. Gallinaceum

Bass and Johns (1915) described a technique for concentrating human malaria parasites for diagnostic and other purposes by the differential centrifugation of physiological saline suspensions of malarial blood. Concentration of Plasmodium knowlesi of Macaca mullata by sedimentation method was found quite satisfactory for making certain biochemical and physiological investigations (Ball, Anfinsen, Geiman, McKee and Ormsbee, 1945; Deane, 1945). Erythrocytes containing Plasmodium vivax have been found to be in general larger and lighter than the normal erythrocytes. Later workers (Bass and Johns, 1915; Kitchen, 1938; Ferrebee, Gibson, and Peacock, 1945; and Ferrebee and Geiman, 1946) showed, that the usual method for concentration of plasmodia by differential sedimentation was unsuitable and they evolved a method using a solution of plasma albumin v/hich gave a partial separation of the normal and infected cells. In our study of the physiology of the malarial parasites and certain immunological investigations it was. essential to obtain concentrates of infected erythrocytes and malarial parasites in the free form. In the present communication a procedure adopted for obtaining concentrates of infected erythrocytes and free malarial parasites has been described.
